Joint efforts, adjustment of the Allium cepa test methodology as support for assessing genotoxicity in the Una River basin and the role of the Lagos São João-RJ Watershed Committee in the planning of monitoring actions

With increased pressure of the multiple uses of water resources and the consequent increase in water pollution, ecotoxicological studies are essential for monitoring water quality in order to manage and restore water bodies. This work aims at carrying out a study that contemplates the partnership with socio-environmental actors and that promotes the planning of ecotoxicological monitoring actions useful for environmental managers. In this way, consultations and meetings were held with members of the Lagos São João Watershed Committee to guide the monitoring proposal, thenleading to the laboratory, where the methodology for applying the Allium cepa (onion) test was carried out for ecotoxicological assessment in the Una River. Within the standardization, different concentrations of Potassium Dichromate (K2Cr2O7) were used and then the presence of indicators of cytotoxic and genotoxic effects were analyzed. Different indicators were found, amongst them the presence of a wandering chromosome, micronucleus, spindle abnormality, chromosomal adhesion, irregular anaphase and anaphase bridge. After adjustments and standardization of the method, it will be used to monitor the points established in the meetings with the representatives of the Lagos São João Watershed Committee.Downloads
